概要: application/pdf<br />Departmental Bulletin Paper<br />The localization of metallothionein (MT), a small molecular weight heavy metal binding protein in the human brain, was investigated by the immunohistochemical technique. The amounts of MT and heavy metals (Zn, Cu, Cd) were assayed by radioimmunoassay and atomic absorption spectrophotometer, respectively. Immunohistochemically, MT was found in the pia mater, ependymal cells, astrocytes and neuropil of the telencephalon. In contrast, oligodendroglia and microglia did not show positive immunostaining for MT. The cytoplasm, processes and some nuclei of astrocytes showed strong MT immunostaining. Vascular feet and adventitia were also postitive for MT immunostaining. Moreover, the pia mater, astrocytes, neuropil and ependyma in the diencephalon, mesencephalon, pons, medulla oblongata and spinal cord showed the same positive immunostaining for MT as the telencephalon. In the cerebellum, Bergmann's glias and astrocytes of the granular layer and white matter showed strongly positivity for the MT immunostain. According to the radioimmunoassay, the amount of MT in the brain was relatively high about 36.0μg/g wet weight. For the heavy metals in the brain, zinc, copper and cadmium were detected. 続きを見る

概要: application/pdf<br />Departmental Bulletin Paper<br />Light microscopically and immunohistochemically, we observed the effects of intravenous administration of copper, zinc, silver, cadmium, gold and mercury on metallothionein (MT) in rabbit tissue. Normal rabbit tissue, except the accessory lacrimal gland, stained positively for MT. Heavy metal treatment induced changes in immunostaining for MT in various tissues, for example cadmium and zinc increased stainability for the MT immunostain in most tissues, but administration of mercury and silver induced decreases in some. 続きを見る

Strongly positive immunostaining for MT was found in the kidney, liver and brain, but the heart and aorta showed only weak immunostaining for this protein. According to the results of radioimmunoassay, the level of MT in the kidney, liver, brain and stomach was relatively high, but that in the heart and aorta was low. As heavy metals in the various organs, Zn, Cu and Cd were detected. The amount of Zn was highest in the liver and that of Cd was highest in the kidney. 続きを見る

概要: application/pdf<br />Departmental Bulletin Paper<br />Immunohistochemically, the localization of metallothionein (MT) was investigated using four antibodies: anti-MT-ascaris anti-MT-bovine serum albumin, anti-MT-polyvinyl pyrrolidone and anti-MT-keyhole limpets hemocyanin. The four antibodies showed positive MT immunostaining, with that of anti-MT-ascaris being the most marked. Immunohistochemically, cardiac muscle, skeletal muscle, endothelial cells, smooth muscel cells, bronchial epithelial cells, bronchial cartilage cells, ducts of salivary glands, esophageal squamous cells, superficial mucous epithelia of the stomach and intestine, Paneth's cells, liver cells, proximal convoluted tubules, transitional cells of the urinary bladder, Seltori cells, the prostate glands, epidermis, were positive for MT immunostaining. 続きを見る
