IL2-treated mice did not. Mice treated with both cyclophosphamide and IL2 survived significantly longer than mice treated with cyclophosphamide alone. IL2 response of lymphocytes was markedly depressed in EL4-bearing mice. Lymphocytes from EL4-bearing mice treated with cyclophosphamide alone responded to IL2 significantly higher than those from mice treated with IL2 alone or those from nontreated mice. Lymphocytes from mice treated with both cyclophosphamide and IL2 responded to IL2 almost same as normal lymphocytes. Cyclophosphamide treatment protected the appearance of both suppressor factor that were found in EL4-bearing mice and inhibited IL2 response of lymphocytes. These results suggested that combination therapy of cyclophosphamide and IL2 showed synergistic antitumor effect through the protection of IL2 responsiveness from suppression by suppressors.

Blastogenic responses against both phytohemagglutinin and pokeweed mitogen of their lymphocytes were markedly suppressed, especially in patients with a higher ratio of OKT8-positive cells. These results suggest the secondary immunodeficiency in hemodialysis patients may be resulted from the combination of the decrease in the lymphocyte number with the depression of lymphocyte activities through the increase in the number of OKT8-positive suppressor cells.

More than 5 times and 3 times the normal activity of PNP were found in B and T lymphocytes, respectively, in S180-bearing mice at 8 weeks after the tumor graft. The S180-bearing mouse T and B lymphocytes with high PNP activity showed suppressor activity against the concanavalin A response of normal lymphocytes. These results suggest that PNP activity in tumor-bearing mice may be related to suppressor cell activity.

anti-B-isohemagglutinins in 117 chronic hemodialysis patients were determined and compared with those in 116 healthy controls. Serum IgM concentration was significantly retarded in the patients as compared with that in the controls, while no remarkable differences were observed between the concentrations of IgG and IgA in the patients' sera and those in healthy controls. Both anti-A- and anti-B-isohemagglutinin titers which are characteristic IgM class antibodies were significantly lower in the patients than those in the controls. Antistreptolysin O titer which is IgG class antibody was also significantly retarded in the hemodialysis patients. These results suggest that chronic hemodialysis may inhibit the production of both IgM and IgG, especially of IgM antibody.

blastogenic responses to phytohemagglutinin P (PHA-P) or bacterial lipopoly saccharide (LPS) and on adenosine deaminase (ADA) or purine nucleoside phosphorylase (PNP) activities were studied with spleen lymphocytes. The retardation of blastogenic responses to both PHA-P and LPS were associated with the retardation of both intracellular ADA and PNP activities in 6MP-R-treated mouse spleen lymphocytes. Both PHA and LPS responses were suppressed in lymphocytes from mice treated with predonine, whereas no suppression of both ADA and PNP activities was observed. In mice treated with CY, LPS response of spleen lymphocytes was markedly suppressed but PHA response was almost the same with that of normal lymphocytes. PNP activity was retarded in lymphocytes of CY-treated mice, while ADA activity of them was rather enahnced. These results suggest that the suppression with 6MP-R may be corresponding to the inherited immunodeficiency with enzyme deficiency, and that the suppression with predonine may be corresponding to immunodeficiency which is not associated with enzyme deficiency.

la typhimurium was examined by antilethal infection immunity, bactericidal activity of macrophages and by histopathological studies of livers of immunized mice. Similar to mice immunized with live whole cells of λ9 strain, mice immunized with mini-cells acquired anti-infection immunity against intraperitoneal challenge with LT_2 strain of S. typhimurium, while no anti-infection immunity was observed in mice immunized with heat-killed whole cells of S. typhimurium. Macrophages collected from mice which had been immunized with mini-cells exhibited enhancing bactericidal activity against Salmonella cells, while no enhancement of bactericidal activity was observed in macrophoges collected from mice which had been immunized with heat-killed whole cells. Moreover, by histopathological study, granuloma formation was observed in the livers of mice which were immunized with mini-cells or live whole cells, but not in those of mice immunized with heat-killed whole cells. These results strongly suggested that mini-cells have immunogenic properties similar to those of live whole cells of S. typhimurium.

was retarded when human peripheral white blood cells (WBC) including sensitized lymphocytes were mixed with the corresponding antigen (PPD, chlorpromazine or cephalexin). By the contrast, the addition of the antigen did not inhibit the migration of PMN leukocytes when the WBC including nonsensitized lymphocytes were used for assay. These results suggest that we are able to assay in vitro delayed hypersensitivity by the migration inhibition test with Boyden's chamber. With our method, more quantitative results can be obtained in a shorter time than other methods.

ng the presence of macrophage chemotaxis inhibitory factor (MCIF). MCIF inhibited chemotaxis not only of syngeneic but also of xenogeneic macrophages through the depression of macrophage adherence. MCIF was heat-labile, and its activity was lost within 3 days at 4℃ but was preserved 2 month at -80℃. MCIF which was assayed with xenogeneic mouse macrophages was found from early stage in sera of human patients with mammary carcinoma. Ia-negative macrophages were more sensitive to MCIF than Ia-positive macrophages suggesting that nonspecific macrophage activities such as chemotactic or digestive activities were at first depressed in tumor-bearing hosts and that the depression of such activities might cause relaps of tumor cells or serious infection in tumor-bearing hosts.
